Activation of ALDH1A1 by omeprazole reduces cell oxidative stress damage

چکیده
Under physiological conditions, cells produce low basal levels of reactive oxygen species (ROS); however, in pathologic conditions ROS production increases dramatically, generating high concentrations toxic unsaturated aldehydes. Aldehyde dehydrogenases (ALDHs) are responsible for detoxification these aldehydes protecting the cell. Due to relevance enzymes, it is important design strategies modulate their activity. It was previously reported that omeprazole activation ALDH1A1 protected Escherichia coli overexpressing this enzyme, from oxidative stress generated by H2O2. In work, cell protection potential evaluated eukaryotic cells. AS-30D or hepatocyte suspensions were subjected a treatment with and exposure light (that required activate active site ALDH) then exposed Cells showed viability similar control cells, total activity ALDH preserved, while lipid markers maintained low. Cell avoided inhibition ALDHs disulfiram, revealing key role enzymes protection. Additionally, also preserved ALDH2 (mitochondrial isoform) activity, diminishing aldehyde organelle, mitochondrial respiration transmembrane formation capacity, These results highlight as part antioxidant system cell, since if decreases under compromised.

Chinese green tea consumption reduces oxidative stress, inflammation and tissues damage in smoke exposed rats
Objective(s):One cause of cigarette smoking is oxidative stress that may alter the cellular antioxidant defense system, induce apoptosis in lung tissue, inflammation and damage in liver, lung, and kidney. It has been shown that Chinese green tea (CGT) (Lung Chen Tea) has higher antioxidant property than black tea. In this paper, we will explore the preventive effect of CGT on cigarette smoke-in...

Aims: In this study, we investigated the effect of clove extract (Syzygium aromaticum) on liver cell damage and oxidative stress caused by diabetes in adult rats. Methods & Materials: For this study, 28 female rats were collected and divided into four groups: A: Control group; B: Diabetic Control group (DC) which received 20% glycerol dissolved in normal saline as carriers; C: Diabetic rats (D...
متن کاملCrocin Reduces Oxidative Stress Produced by Busulfan Administration on Fertility of Male Rats
Background & Objective: Busulfan (BUS) as a drug for cancer treatment can produce free radicals. Crocin (CRO) is a potent antioxidant with beneficial effects. This study was designed to evaluate the effects of CRO against BUS-related damages to the male rats reproductive parameters. Materials & Methods: Forty-eight male rats were randomly assigned into 8 groups; Group 1 (normal control group) ...
متن کاملCytoprotective effect of phloroglucinol on oxidative stress induced cell damage via catalase activation.
We investigated the cytoprotective effect of phloroglucinol, which was isolated from Ecklonia cava (brown alga), against oxidative stress induced cell damage in Chinese hamster lung fibroblast (V79-4) cells. Phloroglucinol was found to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, hydrogen peroxide (H(2)O(2)), hydroxy radical, intracellular reactive oxygen species (ROS), and thus preve...
متن کاملVitamin D3 Reduces Tissue Damage and Oxidative Stress Caused by Exhaustive Exercise
Exhaustive exercise results in inflammation and oxidative stress, which can damage tissue. Previous studies have shown that vitamin D has both anti-inflammatory and antiperoxidative activity. Therefore, we aimed to test if vitamin D could reduce the damage caused by exhaustive exercise. Rats were randomized to one of four groups: control, vitamin D, exercise, and vitamin D+exercise. Exercised r...
